Background:

Glycolytic enzyme that catalyzes the transfer of a phosphoryl group from phosphoenolpyruvate (PEP) to ADP, generating ATP. Stimulates POU5F1-mediated transcriptional activation. Plays a general role in caspase independent cell death of tumor cells. The ratio betwween the highly active tetrameric form and nearly inactive dimeric form determines whether glucose carbons are channeled to biosynthetic processes or used for glycolytic ATP production. The transition between the 2 forms contributes to the control of glycolysis and is important for tumor cell proliferation and survival.

VALIDATION IMAGES

Mouse heart lysates probed with PKM2 Polyclonal Antibody, unconjugated (bs-0101R) at 1:300 overnight at 4°C followed by a conjugated secondary antibody at 1:10000 for 60 minutes at 37°C.


Lane 1: Rat Testis lysates; Lane 2: Rat Prostate lysates; Lane 3: Rat Kidney lysates probed with PKM2 Polyclonal Antibody, Unconjugated (bs-0101R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.


A549 cells were fixed with 4% PFA for 10min at room temperature,permeabilized with 0.1% PBST for 20 min at room temperature, and incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with PKM2 Polyclonal Antibody(bs-0101R)at 1:100 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2%BSA in PBS, followed by secondary antibody incubation for 40 min at room temperature. Acquisitions of 20,000 events were performed. Cells stained with primary antibody (green), and isotype control (orange).
