Background:

AKT1-specific substrates have been recently identified, including palladin (PALLD), which phosphorylation modulates cytoskeletal organization and cell motility; prohibitin (PHB), playing an important role in cell metabolism and proliferation; and CDKN1A, for which phosphorylation at 'Thr-145' induces its release from CDK2 and cytoplasmic relocalization. These recent findings indicate that the AKT1 isoform has a more specific role in cell motility and proliferation. Phosphorylates CLK2 thereby controlling cell survival to ionizing radiation.

VALIDATION IMAGES

Formalin-fixed and paraffin embedded: mouse transplant lymphoma labeled with Anti-PKB Polyclonal Antibody, Unconjugated 1:200 followed by conjugation to the secondary antibody and DAB staining


Lane 1: Mouse Placenta lysates;Lane 2: Mouse Lung lysates probed with AKT1/2/3 Polyclonal Antibody, unconjugated (bs-0115R) at 1:300 overnight at 4°C followed by a conjugated secondary antibody at 1:10000 for 60 minutes at 37°C.


Tissue/cell: A549 cell; 4% Paraformaldehyde-fixed; Triton X-100 at room temperature for 20 min; Blocking buffer (normal goat serum, C-0005) at 37°C for 20 min; Antibody incubation with (AKT1) polyclonal Antibody, Unconjugated (bs-0115R) 1:100, 90 minutes at 37°C; followed by a FITC conjugated Goat Anti-Rabbit IgG antibody at 37°C for 90 minutes, DAPI (blue, C02-04002) was used to stain the cell nuclei.
