Background:
The muscarinic acetylcholine receptor mediates various cellular responses, including inhibition of adenylate cyclase, breakdown of phosphoinositides and modulation of potassium channels through the action of G proteins. Primary transducing effect is adenylate cyclase inhibition. Signaling promotes phospholipase C activity, leading to the release of inositol trisphosphate (IP3); this then triggers calcium ion release into the cytosol.
VALIDATION IMAGES
Mouse brain lysates probed with ChRM2 Polyclonal Antibody, Unconjugated (bs-0236R) at 1:500 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:10000 for 60 min at 37˚C.
Lane 1: Mouse Spinal cord tissue lysates; Lane 2: Mouse Heart tissue lysates; Lane 3: Rat Spinal cord tissue lysates; Lane 4: Rat Heart tissue lysates probed with ChRM2 Polyclonal Antibody, Unconjugated (bs-0236R) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
Paraformaldehyde-fixed, paraffin embedded (Human kidney); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with (ChRM2) Polyclonal Antibody, Unconjugated (bs-0236R) at 1:200 overnight at 4°C, followed by operating according to SP Kit(Rabbit) (sp-0023) instructionsand DAB staining.
