Background:

Important in cell-cell recognition. Appears to function in leukocyte-endothelial cell adhesion. Interacts with integrin alpha-4/beta-1 (ITGA4/ITGB1) on leukocytes, and mediates both adhesion and signal transduction. The VCAM1/ITGA4/ITGB1 interaction may play a pathophysiologic role both in immune responses and in leukocyte emigration to sites of inflammation.

VALIDATION IMAGES

Lane 1: Human VCAM1 protein; Probed with Rabbit Anti-VCAM1 Polyclonal Antibody, Unconjugated (bs-0396R) at 1:300 overnight at 4˚C. Followed by a conjugated secondary antibody at 1:20000 for 90 min at 37˚C.


Mouse spleen cells (dark blue) were fixed with 2% paraformaldehyde for 10 min at 37℃. Primary antibody, Rabbit Anti-VCAM-1 antibody (bs-0396R, Green) at 1μg /8x10^5 cells was incubated for 30 min at room temperature, followed by 1 X PBS containing 0.5% BSA + 1 0% goat serum (1 hour) to block non-specific protein-protein interactions. Then Goat Anti-rabbit IgG/FITC (light blue) antibody was added into the blocking buffer mentioned above to react with the primary antibody at 1/200 dilution for 40 min at room temperature. Isotype control Rabbit IgG (orange) was analyzed under the same conditions.


Lane 1: Human Siha cell lysates; Lane 2: Human HUVEC cell lysates; Lane 3: Human U251 cell lysates probed with VCAM-1 Polyclonal Antibody, Unconjugated (bs-0396R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.


Lane 1: Mouse Kidney tissue lysates; Lane 2: Mouse Spleen tissue lysates; Lane 3: Mouse Cerebrum tissue lysates probed with VCAM1 Polyclonal Antibody, Unconjugated (bs-0396R) at 1:500 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
