Background:

Transcription factor that forms a trimeric complex with OCT4 on DNA and controls the expression of a number of genes involved in embryonic development such as YES1, FGF4, UTF1 and ZFP26. Critical for early embryogenesis and for embryonic stem cell pluripotency. May function as a switch in neuronal development. Downstream SRRT target that mediates the promotion of neural stem cell self-renewal. Keeps neural cells undifferentiated by counteracting the activity of proneural proteins and suppresses neuronal differentiation (By similarity).

VALIDATION IMAGES

NCCIT cells probed with Rabbit Anti-SOX2 Polyclonal Antibody (bs-0523R) at 1:100 for 60 minutes at room temperature followed by Goat Anti-Rabbit IgG (H+L) Alexa Fluor 488 Conjugated Secondary Antibody.


Lane 1: mouse embryo lysates Lane 2: mouse lung lysates probed with Anti SOX2 Polyclonal Antibody, Unconjugated (bs-0523R) at 1:200 in 4˚C. Followed by conjugation to secondary antibody (bs-0295G-HRP) at 1:3000 90min in RT. Predicted band 32kD. Observed band size: 32kD.


Paraformaldehyde-fixed, paraffin embedded Mouse embryo; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with SOX2 Polyclonal Antibody, Unconjugated (bs-0523R) at 1:400 overnight at 4°C, DAB staining.


U251 cells (black) were fixed with 4% PFA for 10min at room temperature,permeabilized with PBST for 20 min at room temperature, and incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with SOX2 Antibody(bs-0523R)at 1:50 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2%BSA in PBS, followed by secondary antibody(blue) incubation for 40 min at room temperature. Acquisitions of 20,000 events were performed. Cells stained with primary antibody (green), and isotype control (orange).
