Background:

Possible adhesion molecule with a role in early hematopoiesis by mediating the attachment of stem cells to the bone marrow extracellular matrix or directly to stromal cells. Could act as a scaffold for the attachment of lineage specific glycans, allowing stem cells to bind to lectins expressed by stromal cells or other marrow components. Presents carbohydrate ligands to selectins.

Formalin-fixed and paraffin embedded human gastric adenocarcinoma labeled Anti-CD34 Polyclonal Antibody, Unconjugated (bs-0646R) at 1:200, followed by conjugation to the secondary antibody Goat Anti-Rabbit IgG, AF488 conjugated(bs-0295G-AF488)used at 1:200 dilution for 40 minutes at 37°C


Mouse lung lysate probed with Anti-CD34 Polyclonal Antibody, Unconjugated (bs-0646R) at 1:300 overnight at 4˚C. Followed by conjugation to secondary antibody (bs-0295G-HRP) at 1:5000 for 90 min at 37˚C.


Mouse heart lysates probed with CD34 Polyclonal Antibody, unconjugated (bs-0646R) at 1:300 overnight at 4°C followed by a conjugated secondary antibody at 1:10000 for 90 minutes at 37°C.


Mouse brain lysates probed with Anti-CD34 Polyclonal Antibody, Unconjugated (bs-0646R) at 1:3200 in 4˚C. Followed by conjugation to secondary antibody (bs-0295G-HRP) at 1:5000 90min in 37˚C.


Human Raji cells probed with CD34 Polyclonal Antibody, Unconjugated (bs-0646R) (green) at 1:100 for 30 minutes followed by a PE conjugated secondary antibody compared to unstained cells (blue), secondary only (light blue), and isotype control (orange).


Paraformaldehyde-fixed, paraffin embedded Human kidney; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with CD34 Polyclonal Antibody, Unconjugated (bs-0646R) at 1:400 overnight at 4°C, followed by a conjugated secondary antibody for 20 minutes and DAB staining.


Lane 1: Huvec cell lysates; Lane 2: Molt-4 cell lysates probed with CD34 Polyclonal Antibody, Unconjugated (bs-0646R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
