Background:

Cooperates with LY96 and CD14 to mediate the innate immune response to bacterial lipopolysaccharide (LPS). Acts via MYD88, TIRAP and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response. Also involved in LPS-independent inflammatory responses triggered by free fatty acids, such as palmitate. In complex with TLR6, promotes sterile inflammation in monocytes/macrophages in response to oxidized low-density lipoprotein (oxLDL) or amyloid-beta 42. In this context, the initial signal is provided by oxLDL- or amyloid-beta 42-binding to CD36. This event induces the formation of a heterodimer of TLR4 and TLR6, which is rapidly internalized and triggers inflammatory response, leading to the NF-kappa-B-dependent production of CXCL1, CXCL2 and CCL9 cytokines, via MYD88 signaling pathway, and CCL5 cytokine, via TICAM1 signaling pathway, as well as IL1B secretion.

Image submitted by One World Lab validation program. HL60 and MCF-7 cells were stained with rabbit polyclonal antibody against TLR4 with two dilutions (1:100 and 1:250). 2nd antibody without primary antibody was used as control included here.


Formalin-fixed and paraffin embedded human placenta labeled with Rabbit Anti TLR4/CD284 Polyclonal Antibody, Unconjugated (bs-1021R) at 1:200 followed by conjugation to the secondary antibody and DAB staining


Formalin-fixed and paraffin embedded rat heart labeled with Rabbit Anti TLR4/CD284 Polyclonal Antibody, Unconjugated (bs-1021R) at 1:200 followed by conjugation to the secondary antibody and DAB staining


Mouse macrophages(RAW264.7)labeled with Rabbit Anti TLR4/CD284 Polyclonal Antibody, Unconjugated (bs-1021R) at 1:200 followed by conjugation to the secondary antibody and DAB staining


Paraformaldehyde-fixed, paraffin embedded Human esophageal carcinoma; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with TLR4 Polyclonal Antibody, Unconjugated (bs-1021R) at 1:200 overnight at 4°C, DAB staining.


K562 cells were incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with TLR4 Polyclonal Antibody(bs-1021R)at 1:50 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2%BSA in PBS, followed by secondary antibody incubation for 40 min at room temperature. Acquisitions of 20,000 events were performed. Cells stained with primary antibody (green), and isotype control (orange).


Lane 1: Human MDA-MB-231 cell lysates; Lane 2: Human HUVEC cell lysates; Lane 3: Human HL-60 cell lysates probed with TLR4 Polyclonal Antibody, Unconjugated (bs-1021R) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
