Background:

Coagulation factor II (thrombin) receptor-like 1 (F2RL1) is a member of the large family of 7-transmembrane-region receptors that couple to guanosine-nucleotide-binding proteins. F2RL1 is also a member of the protease-activated receptor family. It is activated by trypsin, but not by thrombin. It is activated by proteolytic cleavage of its extracellular amino terminus. The new amino terminus functions as a tethered ligand and activates the receptor. The F2RL1 gene contains two exons and is widely expressed in human tissues. The predicted protein sequence is 83% identical to the mouse receptor sequence. [provided by RefSeq, Jul 2008]

VALIDATION IMAGES

Paraformaldehyde-fixed, paraffin embedded human colon cancer; Antigen retrieval by boiling in sodium citrate buffer (pH6) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 30 minutes; Blocking buffer (normal goat serum) at 37°C for 20min; Antibody incubation with Rabbit Anti-PAR-2 Polyclonal Antibody, Unconjugated (bs-1178R) at 1:500 overnight at 4°C, followed by a conjugated secondary and DAB staining.


Lane 1: mouse brain lysates Lane 2: mouse embryo lysates probed with Anti PAR-2 Polyclonal Antibody, Unconjugated (bs-1178R) at 1:200 in 4˚C. Followed by conjugation to secondary antibody (bs-0295G-HRP) at 1:3000 90min in RT. Predicted band 40kD. Observed band size: 40kD.


Formalin-fixed and paraffin embedded rat colitis tissue labeled with Anti-PAR-2 Polyclonal Antibody, Unconjugated (bs-1178R) at 1:200 followed by conjugation to the secondary antibody and DAB staining


Lane 1: HepG2 cell lysates; Lane 2: MCF-7 cell lysates probed with PAR-2 Polyclonal Antibody, Unconjugated (bs-1178R) at 1:500 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
