Background:

This enzyme is necessary for target cell lysis in cell-mediated immune responses. It cleaves after Asp. Seems to be linked to an activation cascade of caspases (aspartate-specific cysteine proteases) responsible for apoptosis execution. Cleaves caspase-3, -7, -9 and 10 to give rise to active enzymes mediating apoptosis.

Formalin-fixed and paraffin embedded human lung carcinoma labeled with Anti-Granzyme B Polyclonal Antibody, Unconjugated (bs-1351R) at 1:200 followed by conjugation to the secondary antibody and DAB staining


Antigen: bs-1351P, 0.2ug/100ul \nPrimary: Antiserum, 1:500, 1:1000, 1:2000, 1:4000, 1:8000, 1:16000, 1:32000; \nSecondary: HRP conjugated Goat Anti-Rabbit IgG(bs-0295G-HRP) at 1: 5000; \nTMB staining; Read the data in MicroplateReader by 450nm. \n


U-937 cells were fixed with 4% PFA for 10min at room temperature,permeabilized with 0.1% PBST for 20 min at room temperature, and incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with Granzyme B Antibody(bs-1351R)at 1:50 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2%BSA in PBS, followed by secondary antibody incubation for 40 min at room temperature. Acquisitions of 20,000 events were performed. Cells stained with primary antibody (green), and isotype control (orange).


Lane 1: Human K562 cell lysates; Lane 2: Human MOLT4 cell lysates probed with Granzyme B Polyclonal Antibody, Unconjugated (bs-1351R) at 1:500 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at room temperature for 60 min.
