Background:

Claudins function as major constituents of the tight junction complexes that regulate the permeability of epithelia. While some claudin family members play essential roles in the formation of impermeable barriers, others mediate the permeability to ions and small molecules. Often, several claudin family members are coexpressed and interact with each other, and this determines the overall permeability. CLDN1 is required to prevent the paracellular diffusion of small molecules through tight junctions in the epidermis and is required for the normal barrier function of the skin. Required for normal water homeostasis and to prevent excessive water loss through the skin, probably via an indirect effect on the expression levels of other proteins, since CLDN1 itself seems to be dispensable for water barrier formation in keratinocyte tight junctions.

VALIDATION IMAGES

HeLa lysates probed with Rabbit Anti-Claudin 1 Polyclonal Antibody (bs-1428R) at 1:300 overnight at 4˚C. Followed by conjugation to secondary antibody (bs-0295G-HRP) at 1:5000 for 90 min at 37˚C.


Paraformaldehyde-fixed, paraffin embedded rat bladder; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with Claudin 1 Polyclonal Antibody, Unconjugated (bs-1428R) at 1:400 overnight at 4°C, followed by a conjugated secondary for 20 minutes and DAB staining.


Lane 1: Human A549 cell lysates; Lane 2: Human HepG2 cell lysates probed with Claudin 1 Polyclonal Antibody, Unconjugated (bs-1428R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
