Background:

Cd177 (NP1), a glycosyl phosphatidylinositol (GPI) linked N glycosylated cell surface glycoprotein, was first described in a case of neonatal alloimmune neutropenia and analyses showed cell surface expression of a 60 kDa protein, 14 kDa greater than the predicted size, probably due to the presence of 3 potential N glycosylation sites. Immunohistochemistry demonstrated expression in bone marrow early erythroblasts, megakaryocytes, promyelocytes, and myelocytes. The neutrophil NB1 antigen (CD177) is expressed by 97% of the caucasian population. against NB1 can be involved in neonatal alloimmune neutropenia (NAN).

VALIDATION IMAGES

Formalin-fixed and paraffin embedded rat colon tissue labeled with Anti CD177/NB1 Polyclonal Antibody, Unconjugated (bs-1482R) at 1:200 followed by conjugation to the secondary antibody and DAB staining.


Lane 1: Hela Cells; Lane 2: K562 Cells; Lane 3: MCF-7 Cells; Lane 4: 293T Cells; Lane 5: Sp2/0 Cells; Lane 6: NIH/3T3 Cells; 40ug of lysate loaded into each lane; Probed with CD177 Polyclonal Antibody, unconjugated (bs-1482R) at 1:1000 overnight at 4°C followed by a conjugated secondary antibody for 60 minutes at 37°C.


Lane 1: Mouse Spleen lysates; Lane 2: Mouse Raw264.7 cell lysates; Lane 3: Rat Lymph node lysates probed with CD177 Polyclonal Antibody, Unconjugated (bs-1482R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
