Background:

The protein encoded by this gene belongs to the Ser/Thr protein kinase family. It is required for checkpoint mediated cell cycle arrest in response to DNA damage or the presence of unreplicated DNA. This protein acts to integrate signals from ATM and ATR, two cell cycle proteins involved in DNA damage responses, that also associate with chromatin in meiotic prophase I. Phosphorylation of CDC25A protein phosphatase by this protein is required for cells to delay cell cycle progression in response to double-strand DNA breaks. Several alternatively spliced transcript variants have been found for this gene. [provided by RefSeq, Oct 2011].

Lane 1: rat brain lysates Lane 2: rat heart lysates probed with Anti CHK1/CHEK1 Polyclonal Antibody, Unconjugated (bs-1681R) at 1:200 in 4˚C. Followed by conjugation to secondary antibody (bs-0295G-HRP) at 1:3000 90min in RT. Predicted band 52kD. Observed band size: 33kD, 52kD.


Paraformaldehyde-fixed, paraffin embedded Human brain glioma; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with CHK1/CHEK1 Polyclonal Antibody, Unconjugated (bs-1681R) at 1:400 overnight at 4°C, DAB staining.
