Background:

Heme oxygenase cleaves the heme ring at the alpha methene bridge to form biliverdin. Biliverdin is subsequently converted to bilirubin by biliverdin reductase. Under physiological conditions, the activity of heme oxygenase is highest in the spleen, where senescent erythrocytes are sequestrated and destroyed. Exhibits cytoprotective effects since excess of free heme sensitizes cells to undergo apoptosis.

VALIDATION IMAGES

Paraformaldehyde-fixed, paraffin embedded rat lung tissue; Antigen retrieval by boiling in sodium citrate buffer(pH6) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 30 minutes; Blocking buffer (normal goat serum) at 37°C for 20min; Antibody incubation with Rabbit Anti-Heme Oxygenase Polyclonal Antibody, Unconjugated (bs-2075) at 1:500 overnight at 4°C, followed by a conjugated secondary and DAB staining


293T cell lysate probed (bs-2075R) at 1:300 overnight in 4˚C. Followed by conjugation to the secondary antibody (bs-0295G-HRP) at 1:5000 90min in 37˚C. \n


Lane 1: Human HepG2 cell lysates; Lane 2: Human A549 cell lysates; Lane 3: Human DU145 cell lysates; Lane 4: Human H9C2 cell lysates; Lane 5: Human 293T cell lysates probed with Heme Oxygenase Polyclonal Antibody, Unconjugated (bs-2075R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
