Background:

Component of a Polycomb group (PcG) multiprotein PRC1-like complex, a complex class required to maintain the transcriptionally repressive state of many genes, including Hox genes, throughout development. PcG PRC1 complex acts via chromatin remodeling and modification of histones; it mediates monoubiquitination of histone H2A 'Lys-119', rendering chromatin heritably changed in its expressibility. In the PRC1 complex, it is required to stimulate the E3 ubiquitin-protein ligase activity of RNF2/RING2.

Formalin-fixed and paraffin embedded human colon carcinoma labeled with Rabbit Anti BMI1/PCGF4 Polyclonal Antibody, Unconjugated (bs-2999R) at 1:200 followed by conjugation to the secondary antibody and DAB staining

Lane 1: Mouse NIH/3T3 cell lysates; Lane 2: Human MCF-7 cell lysates probed with PCGF4/BMI1 Polyclonal Antibody, Unconjugated (bs-2999R) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.


NIH/3T3 cells were fixed with 4% PFA for 10min at room temperature , permeabilized with 90% ice-cold methanol for 20 min at -20℃, and incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with PCGF4/BMI1 Polyclonal Antibody(bs-2999R)at 1:100 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2%BSA in PBS, followed by secondary antibody incubation for 40 min at room temperature. Acquisitions of 20,000 events were performed. Cells stained with primary antibody (green), and isotype control (orange).
