Background:

Members of the 14-3-3 family of proteins are highly conserved proteins, localized in neurons, and are axonally transported to the nerve terminals. They are also present, at lower levels, in various other eukaryotic tissues. 14-3-3 proteins appear to play important roles in a variety of signal transduction pathways, including those involved in cell cycle regulation and cell survival. Because 14-3-3 proteins bind to specific phosphoserine-containing sequences they are likely to have an important role in signaling pathways mediated by serine/threonine protein kinases. Evidence indicates 14-3-3 is required for Raf 1 kinase activity and phosphorylation amoung many other functions.

VALIDATION IMAGES

HepG2 cell lysates probed with 14-3-3 protein zeta (Ser58) Polyclonal Antibody, Unconjugated (bs-3000R) at 1:300 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.


A549 cells(black) were fixed with 4% PFA for 10min at room temperature,permeabilized with 0.1% PBST for 20 min at room temperature, and incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with 14-3-3 zeta / delta (Ser58) Polyclonal Antibody(bs-3000R)at 1:50 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2% BSA in PBS, followed by secondary antibody(blue) incubation for 40 min at room temperature. Acquisitions of 20,000 events were performed. Cells stained with primary antibody (green), and isotype control (orange).
