Background:
This protein tyrosine kinase transmembrane receptor is the receptor for colony stimulating factor 1, a cytokine which controls the production, differentiation, and function of macrophages. This receptor mediates most if not all of the biological effects of this cytokine. Ligand binding activates the receptor kinase through a process of oligomerization and transphosphorylation. The encoded protein is a member of the CSF1/PDGF receptor family of tyrosine protein kinases and contains 5 immunoglobulin like C2 type domains. CD115 is expressed by cells of the monocytic lineage and by progenitor cells. Mutations in this gene have been associated with a predisposition to myeloid malignancy.
VALIDATION IMAGES
Paraformaldehyde-fixed, paraffin embedded mouse brain; Antigen retrieval by boiling in sodium citrate buffer (pH6) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 30 minutes; Blocking buffer (normal goat serum) at 37°C for 20min; Antibody incubation with Rabbit Anti-M-CSF Receptor(Tyr546) Polyclonal Antibody, Unconjugated (bs-3074R) at 1:500 overnight at 4°C, followed by a conjugated secondary and DAB staining.
Lane 1: Human U-87 MG cell lysates; Lane 2: Human A431 cell lysates probed with phospho-MCSF Receptor (Tyr546) Polyclonal Antibody, Unconjugated (bs-3074R) at 1:300 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
