Background:

Variant histone H3 which replaces conventional H3 in a wide range of nucleosomes in active genes. Constitutes the predominant form of histone H3 in non-dividing cells and is incorporated into chromatin independently of DNA synthesis. Deposited at sites of nucleosomal displacement throughout transcribed genes, suggesting that it represents an epigenetic imprint of transcriptionally active chromatin. Nucleosomes wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries which require DNA as a template. Histones thereby play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling.

VALIDATION IMAGES

Formalin-fixed and paraffin embedded mouse brain labeled with Rabbit Anti-Histone H3 (Di Methyl K9) Polyclonal Antibody, Unconjugated (bs-3772R) at 1:200 followed by conjugation to the secondary antibody and DAB staining\n


HeLa cells were fixed with 70% ice-cold methanol overnight at 4℃，permeabilized with 90% ice-cold methanol for 20 min at -20℃, and incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with Histone H3 (Di Methyl K9) Antibody(bs-3772R) at 1：50dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2%BSA in PBS, followed by secondary antibody incubation for 40 min at room temperature. Acquisitions of 20,000 events were performed. Cells stained with primary antibody (green), no antibody control control (blue), secondary antibody only (light blue) and isotype control (orange).
