Background:

Component of the Set1/Ash2 histone methyltransferase (HMT) complex, a complex that specifically methylates 'Lys-4' of histone H3, but not if the neighboring 'Lys-9' residue is already methylated. As part of the MLL1/MLL complex it is involved in methylation and dimethylation at 'Lys-4' of histone H3. May function as a transcriptional regulator. May play a role in hematopoiesis.

VALIDATION IMAGES

NIH3T3 cells were stained with the Ash2 antibody (bs-53055R) and with DAPI. Cells were fixed with 4% formaldehyde for 10’ and blocked with PBS/TX-100 containing 5% normal goat serum and 1% BSA. The cells were immunofluorescently labelled with the Ash2 antibody (left) diluted 1:200 in blocking solution followed by an anti-rabbit antibody conjugated to Alexa488. The middle panel shows staining of the nuclei with DAPI. A merge of the two is shown on the right.


To determine the titer, an ELISA was performed using a serial dilution of the Ash2 antibody (bs-53055R). The wells were coated with the peptides used for immunization of the host rabbit. By plotting the absorbance against the antibody dilution, the titer of the antibody was estimated to be 1:24,000.
