Background:

The protein encoded by this gene catalyzes the post-translational formation of 4-hydroxyproline in hypoxia-inducible factor (HIF) alpha proteins. HIF is a transcriptional complex that plays a central role in mammalian oxygen homeostasis. This protein functions as a cellular oxygen sensor, and under normal oxygen concentration, modification by prolyl hydroxylation is a key regulatory event that targets HIF subunits for proteasomal destruction via the von Hippel-Lindau ubiquitylation complex. Mutations in this gene are associated with erythrocytosis familial type 3 (ECYT3).

L929 cell; 4% Paraformaldehyde-fixed; Triton X-100 at room temperature for 20 min; Blocking buffer (normal goat serum, C-0005) at 37°C for 20 min; Antibody incubation with (EGLN1) polyclonal Antibody, Unconjugated (bs-55063R) 1:100, 90 minutes at 37°C; followed by a conjugated Goat Anti-Rabbit IgG antibody at 37°C for 90 minutes, DAPI (blue, C02-04002) was used to stain the cell nuclei.


Lane 1: 293T cell lysates; Lane 2: EGLN1 knockout (KO) 293T cell lysates probed with EGLN1 Polyclonal Antibody, Unconjugated (bs-55063R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.


Lane 1: Mouse Heart lysates; Lane 2: Mouse Liver lysates; Lane 3: Rat Heart lysates probed with EGLN1 Polyclonal Antibody, Unconjugated (bs-55063R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.


Lane 1: U-87MG cell lysates; Lane 2: 293T cell lysates probed with EGLN1 Polyclonal Antibody, Unconjugated (bs-55063R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
