Background:
The protein encoded by this gene is a member of the protein tyrosine phosphatase (PTP) family. PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. This PTP contains two tandem Src homology-2 domains, which function as phospho-tyrosine binding domains and mediate the interaction of this PTP with its substrates. This PTP is widely expressed in most tissues and plays a regulatory role in various cell signaling events that are important for a diversity of cell functions, such as mitogenic activation, metabolic control, transcription regulation, and cell migration. Mutations in this gene are a cause of Noonan syndrome as well as acute myeloid leukemia. Two transcript variants encoding different isoforms have been found for this gene.
VALIDATION IMAGES
C6 cell; 4% Paraformaldehyde-fixed; Triton X-100 at room temperature for 20 min; Blocking buffer (normal goat serum, C-0005) at 37°C for 20 min; Antibody incubation with (PTPN11) polyclonal Antibody, Unconjugated (bs-55183R) 1:100, 90 minutes at 37°C; followed by a conjugated Goat Anti-Rabbit IgG antibody at 37°C for 90 minutes, DAPI (blue, C02-04002) was used to stain the cell nuclei.
Lane 1: 293T cell lysates; Lane 2: PTPN11 knockout (KO) 293T cell lysates probed with PTPN11 Polyclonal Antibody, Unconjugated (bs-55183R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
Lane 1: U-87MG cell lysates; Lane 2: Jurkat cell lysates; Lane 3: A431 cell lysates; Lane 4: MCF-7 cell lysates; Lane 5: Mouse Brain lysates; Lane 6: Rat Brain lysates; Lane 7: Rat Lung lysates probed with PTPN11 Polyclonal Antibody, Unconjugated (bs-55183R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
