Background:

NAD-dependent protein deacetylase. Activates or deactivates mitochondrial target proteins by deacetylating key lysine residues. Known targets include ACSS1, IDH, GDH, SOD2, PDHA1, LCAD, SDHA and the ATP synthase subunit ATP5O. Contributes to the regulation of the cellular energy metabolism. Important for regulating tissue-specific ATP levels.

Formalin-fixed and paraffin embedded human breast cancer labeled with Anti SIRT3 Polyclonal Antibody, Unconjugated (bs-6105R) followed by conjugation to the secondary antibody and DAB staining


Lane 1: Mouse Liver lysates; Lane 2: Mouse Kindey lysates; Lane 3: Mouse Cerebrum lysates; Lane 4: MouseHeart lysates probed with SIRT3 Polyclonal Antibody, Unconjugated (bs-6105R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.


Paraformaldehyde-fixed, paraffin embedded (rat stomach); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with (SIRT3) Polyclonal Antibody, Unconjugated (bs-6105R) at 1:200 overnight at 4°C, followed by operating according to SP Kit(Rabbit) (sp-0023) instructionsand DAB staining.
