Background:

Receptor for TNFSF6/FASLG. The adapter molecule FADD recruits caspase-8 to the activated receptor. The resulting death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation which initiates the subsequent cascade of caspases (aspartate-specific cysteine proteases) mediating apoptosis. FAS-mediated apoptosis may have a role in the induction of peripheral tolerance, in the antigen-stimulated suicide of mature T-cells, or both. The secreted isoforms 2 to 6 block apoptosis (in vitro).

Lane 1: Rat Serum lysates; Lane 2: Rat Plasma lysates probed with Apo-1 Polyclonal Antibody, Unconjugated (bs-6477R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.


Lane 1: Mouse Thymus lysates; Lane 2: Rat Thymus lysates; Lane 3: Human HepG2 cell lysates; Lane 4: Human Raji cell lysates; Lane 5: Human HT1080 cell lysates; Lane 6: Human A431 cell lysates probed with Apo-1 Polyclonal Antibody, Unconjugated (bs-6477R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.


Raji cells were incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with CD95/FAS Polyclonal Antibody(bs-6477R)at 1:50 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2%BSA in PBS, followed by secondary antibody incubation for 40 min at room temperature. Acquisitions of 20,000 events were performed. Cells stained with primary antibody (green), and isotype control (orange).
