Background:

The protein encoded by this gene is a small cellular or pericellular matrix proteoglycan that is closely related in structure to two other small proteoglycans, decorin and fibromodulin. The encoded protein and decorin are thought to be the result of a gene duplication. Decorin contains one attached glycosaminoglycan chain, while this protein probably contains two chains. For this reason, this protein is called biglycan. This protein plays a role in assembly of collagen fibrils and muscle regeneration. It interacts with several proteins involved in muscular dystrophy, including alpha-dystroglycan, alpha- and gamma-sarcoglycan and collagen VI, and it is critical for the assembly of the dystrophin-associated protein complex.

VALIDATION IMAGES

Paraformaldehyde-fixed (4%) Human Endothelial Cells were blocked with normal goat serum for 20mins at 37℃. The cells were then labeled with Biglycan Polyclonal Antibody, Unconjugated (bs-7552R) at 1:50, overnight at 4°C. Secondary antibody incubation was performed with Goat Anti-Rabbit IgG, Rhodamine B conjugated antibody (bs-0295G-RBITC) at a 1:200 dilution for 40 minutes at 37°C. DAPI (5ug/ml; blue) was used to stain the cell nuclei


Formalin-fixed and paraffin-embedded Human Lung Carcinoma labeled with Biglycan Polyclonal Antibody, Unconjugated (bs-7552R) at 1:200 followed by secondary antibody, DAB and counterstaining


Paraformaldehyde-fixed, paraffin embedded mouse brain; Antigen retrieval by boiling in sodium citrate buffer (pH6) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 30 minutes; Blocking buffer (normal goat serum) at 37°C for 20min; Antibody incubation with Biglycan Polyclonal Antibody (bs-7552R) at 1:400 overnight at 4°C, followed by a conjugated secondary and DAB staining.


Mouse Bone probed with Biglycan Polyclonal Antibody (bs-7552R) at 1:300, overnight at 4˚C. Followed by incubation with a conjugated secondary antibody.
