Background:

This gene encodes a cytokine that functions in inflammation and the maturation of B cells. In addition, the encoded protein has been shown to be an endogenous pyrogen capable of inducing fever in people with autoimmune diseases or infections. The protein is primarily produced at sites of acute and chronic inflammation, where it is secreted into the serum and induces a transcriptional inflammatory response through interleukin 6 receptor, alpha. The functioning of this gene is implicated in a wide variety of inflammation-associated disease states, including suspectibility to diabetes mellitus and systemic juvenile rheumatoid arthritis. VALIDATION IMAGES

Paraformaldehyde-fixed, paraffin embedded Human liver cancer; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with IL-6 (2C6) Monoclonal Antibody, Unconjugated (bsm-10807M) at 1:400 overnight at 4°C, DAB staining.


HUVEC (H) cells were treated with or without LPS (0.5 μg/ml，24h) and Brefeldin A (300 ng/ml，20h) , 25 μg total protein per lane of cell lysates (see on figure) probed with human IL-6 monoclonal antibody, unconjugated (bsm-10807M) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at r.t. for 60 min.
