Background:

Osteocalcin belongs to the osteocalcin/matrix Gla protein family and constitutes 1 to 2% of the total bone protein. It is a 49 amino acid single chain vitamin K dependent protein, made by osteoblasts, and is a major component of the noncollagenous bone matrix. Post translational modification by a vitamin K dependent carboxylase produces three gamma carboxyglutamic acid residues at positions 17, 21 and 24, giving it a high affinity for calcium. It also binds strongly to apatite.

VALIDATION IMAGES

Paraformaldehyde-fixed, paraffin embedded (human skeletal muscle); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with (Osteocalcin) Monoclonal Antibody, Unconjugated (bsm-43106M) at 1:200 overnight at 4°C, followed by operating according to SP Kit(Mouse)(sp-0024) instructionsand DAB staining.


Lane 1: Recombinant human Osteocalcin, C-hFc (HEK293)( bs-43106P) probed with Osteocalcin Monoclonal Antibody, Unconjugated (bsm-43106M) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
