Background:

The major proteolytic product p15 BID allows the release of cytochrome c (By similarity). Isoform 1, isoform 2 and isoform 4 induce ICE-like proteases and apoptosis. Isoform 3 does not induce apoptosis. Counters the protective effect of Bcl-2.

VALIDATION IMAGES

Paraformaldehyde-fixed, paraffin embedded Human lung carcinoma tissue; Antigen retrieval by boiling in sodium citrate buffer(pH6) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 30 minutes; Blocking buffer (normal goat serum) at 37°C for 20min; Antibody incubation with Bid (8D2) monoclonal Antibody, Unconjugated (bsm-50264M) at 1:500 overnight at 4°C, followed by a conjugated secondary and DAB staining


Lane 1 : HeLa Cell Lysate; Lane 2 : A431 Cell Lysate; Lane 3 : K562 Cell Lysate ; Lane 4 : NCI-H460 Cell Lysate; probed with Bid (8D2) Monoclonal Antibody, unconjugated (bsm-50264M) at 1:20000 overnight at 4°C followed by a conjugated secondary antibody at 1:5000 for 60 minutes at Room Temperature.


Immunoprecipitation of A431 cell lysates using Bid (8D2) Monoclonal Antibody (bsm-50264M): Lane 1 : A431 Cell Lysate; Lane 2 : Precipitated from 200ug of Cell Lysate using 2ug of antibody; Lane 3 : Precipitated from 200ug of Cell Lysate using 5ug of antibody; Lane 4 : Precipitated from PBS using 5ug of antibody. WB analysis was performed using bsm-50264M at 0.1ug/mL (1:10000) and incubated at 4℃ overnight, followed secondary antibody incubation for 60min at Room Temperature.
