Source: This ALDH1A1 monoclonal antibody is generated from mouse immunized with ALDH1A1 recombinant protein.

Purification: Purified monoclonal antibody supplied in PBS with 0.09% (W/V) sodium azide. This antibody is purified through a protein G column, eluted with high and low pH buffers and neutralized immediately, followed by dialysis against PBS.

Storage Buffer: 0.01M TBS(pH7.4) with 1% BSA, 0.02% Proclin300 and 50% Glycerol.

Storage: Store at -20°C for 12 months.

Background:

ALDH1A1 belongs to the aldehyde dehydrogenase family. Aldehyde dehydrogenase is the next enzyme after alcohol dehydrogenase in the major pathway of alcohol metabolism. There are two major aldehyde dehydrogenase isozymes in the liver, cytosolic and mitochondrial, which are encoded by distinct genes, and can be distinguished by their electrophoretic mobility, kinetic properties, and subcellular localization. ALDH1A1 is the cytosolic isozyme. Studies in mice show that through its role in retinol metabolism, this gene may also be involved in the regulation of the metabolic responses to high-fat diet. ALDH1A1 binds free retinal and cellular retinol-binding protein-bound retinal. ALDH1A1 can convert/oxidize retinaldehyde to retinoic acid. ALDH1A1 may have a broader specificity and oxidize other aldehydes in vivo.

Lane 1: HepG2 Cell lysates; Lane 2: K562 Cell lysates; Lane 3: NCI-H460 Cell lysates; probed with ALDH1A1 (2C2) Monoclonal Antibody, unconjugated (bsm-51032M) at 1:1000 overnight at 4°C followed by a conjugated secondary antibody for 60 minutes at 37°C.


Lane 1: NCI-H460 Cells; 15 ug of protein lysate loaded; Probed with ALDH1A1 (2C2) Monoclonal Antibody (bsm-51032M) at 1:1000 overnight at 4°C followed by a conjugated secondary antibody for 60 minutes at 37°C.


Paraformaldehyde-fixed and paraffin-embedded Human Colon Cancer tissue incubated with ALDH1A1 (2C2) Monoclonal Antibody (bsm-51032M) at 1:100, overnight at 4°C, followed by a conjugated secondary antibody and DAB staining. Counterstained with hematoxylin.


IF(ICC) staining with ALDH1A1 (2C2) Monoclonal Antibody (bsm-51032M) at 1:25 in MCF-7 cells (green). The nuclear counterstain is DAPI (blue). Cytoplasmic actin is detected with Dylight® 554 Phalloidin (red). Cells were fixed in 4% paraformaldehyde, permeabilized with 0.1% Triton X100/PBS.


Flow cytometric analysis of A549 cells with ALDH1A1 (2C2) Monoclonal Antibody (bsm-51032M) at 1:25 dilution (green) compared with an Isotype control (blue). The cells were fixed with 2% paraformaldehyde (10 min) and then permeabilized with 90% methanol for 10 min. The cells were then incubated in 2% BSA to block non-specific protein-protein interactions. Primary antibody incubation was performed for 60 min at 37ºC.
