Background:

Cysteine protease required for the cytoplasm to vacuole transport (Cvt) and autophagy. Cleaves the C-terminal amino acid of ATG8 family proteins to reveal a C-terminal glycine. Exposure of the glycine at the C-terminus is essential for ATG8 proteins conjugation to phosphatidylethanolamine (PE) and insertion to membranes, which is necessary for autophagy. Preferred substrate is GABARAPL2 followed by MAP1LC3A and GABARAP. Has also an activity of delipidating enzyme for the PE-conjugated forms.

VALIDATION IMAGES

Immunofluorescent analysis of 4% paraformaldehyde-fixed, 0.1% Triton X-100 permeabilized HeLa cells labeled with ATG4A (1458CT808.66.25.69) Monoclonal Antibody (bsm-51292M) at 1/25 dilution, followed by secondary antibody (green), cytoplasmic actin staining (red), and nuclear counter stain with DAPI (blue).


Paraformaldehyde-fixed, paraffin embedded Human brain tissue; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 15 minutes; Blocking buffer (3% BSA) at room temperature for 30min; Antibody incubation with ATG4A (1458CT808.66.25.69) Monoclonal Antibody (bsm-51292M) at 1:25 for 1 hour at 37°C, followed by a conjugated secondary antibody for 20 minutes and DAB staining.


K562 lysates probed with ATG4A (14C9) Monoclonal Antibody, Unconjugated (bsm-51292M) at 1:300 overnight at 4˚C. Followed by a conjugated secondary antibody (bs-0295G-HRP) at 1:5000 for 90 min at 37˚C.
