Background:

May be involved in the translation of target mRNAs by scanning and recognition of the initiation codon. Involved in translation initiation; promotes recruitmnet of aminoacetyled initiator tRNA to P site of 40S ribosomes. Can promote release of deacylated tRNA and mRNA from recycled 40S subunits following ABCE1-mediated dissociation of post-termination ribosomal complexes into subunits. Plays a role in the modulation of the translational profile of a subset of cancer-related mRNAs when recruited to the translational initiation complex by the oncogene MCTS1.

VALIDATION IMAGES

Immunofluorescent analysis of 4% paraformaldehyde-fixed, 0.1% Triton X-100 permeabilized HeLa cells labeled with DENR (2C9) Monoclonal Antibody (bsm-51318M) at 1/25 dilution, followed by secondary antibody (green), cytoplasmic actin staining (red), and nuclear counter stain with DAPI (blue).


Paraformaldehyde-fixed, paraffin embedded human skeletal muscle tissue; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 15 minutes; Blocking buffer (3% BSA) at room temperature for 30min; Antibody incubation with DENR (2C9) Monoclonal Antibody (bsm-51318M) at 1:25 for 1 hour at 37°C, followed by a conjugated secondary antibody for 20 minutes and DAB staining.


Lane 1: A431; Lane 2: human skeletal muscle lysates; Lane 3: human brain Cell lysates; probed with DENR (2C9) Polyclonal Antibody, unconjugated (bsm-51318M) at 1:1000 overnight at 4°C followed by a conjugated secondary antibody for 60 minutes at 37°C.


Hela cells probed with using DENR (2C9) Monoclonal Antibody bsm-51318M at 1:25 for 60 minutes at 37ºC followed by secondary antibody at 1:400 (green), compared to an isotype control of mouse IgG2b (blue).
