Background:

Complexes with metalloproteinases (such as collagenases) and irreversibly inactivates them by binding to their catalytic zinc cofactor. Known to act on MMP-1, MMP-2, MMP-3, MMP-7, MMP-8, MMP-9, MMP-10, MMP-13, MMP-14, MMP-15, MMP-16 and MMP-19.

VALIDATION IMAGES

Lane 1: SW480 Cell lysates; probed with TIMP2 (4C7) Monoclonal Antibody, unconjugated (bsm-51320M) at 1:500 overnight at 4°C followed by a conjugated secondary antibody for 60 minutes at 37°C.


Immunofluorescent analysis of 4% paraformaldehyde-fixed, 0.1% Triton X-100 permeabilized A549 cells labeled with TIMP2 (4C7) Monoclonal Antibody (bsm-51320M) at 1/25 dilution followed by secondary antibody (green), cytoplasmic actin staining (red), and nuclear counter stain with DAPI (blue).


K562 cells probed with using TIMP2 (4C7) Monoclonal Antibody bsm-51320M at 1:25 for 30 minutes followed by A488 secondary antibody (green) at 1:200, compared to an isotype control of mouse IgG1 (blue).


Paraformaldehyde-fixed, paraffin embedded Human kidney tissue; Blocked with 3% BSA for 30min at RT; Antibody incubation with TIMP2 (4C7) Monoclonal Antibody, Unconjugated (bsm-51320M ) at 1:25 for 1hr at 37°C, followed by a conjugated secondary antibody and DAB staining.


Lane 1: Human HT-1080 cell lysates probed with TIMP-2 Monoclonal Antibody, Unconjugated (bsm-51320M) at 1:2000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.

A549 cell; 4% Paraformaldehyde-fixed; Triton X-100 at room temperature for 20 min; Blocking buffer (normal goat serum, C-0005) at 37°C for 20 min; Antibody incubation with (TIMP-2) monoclonal Antibody, Unconjugated (bsm-51320M) 1:25, 90 minutes at 37°C; followed by a conjugated Goat Anti-Mouse IgG antibody at 37°C for 90 minutes, DAPI (blue, C02-04002) was used to stain the cell nuclei.


K562 cells were fixed with 2% PFA (10min at room temperature)and then permeabilized with 90% methanol for 10 min.The cells were then incubated in 2%BSA to block non-specific protein-protein interactions for 30 min at room temperature .Cells stained with Primary Antibody for 30 min at room temperature. The secondary antibody used for 40 min at room temperature. Acquisition of 20,000 events was performed.
