Source: This CAPN2 antibody is generated from a mouse immunized with a KLH conjugated synthetic peptide between amino acids from the human region of human CAPN2.

Purification: Purified monoclonal antibody supplied in PBS with 0.09% (W/V) sodium azide. This antibody is purified through a protein G column, followed by dialysis against PBS.

Storage Buffer: 0.01M TBS(pH7.4) with 1% BSA, 0.02% Proclin300 and 50% Glycerol.

Storage: Store at -20°C for 12 months.

Background:

Calcium-regulated non-lysosomal thiol-protease which catalyze limited proteolysis of substrates involved in cytoskeletal remodeling and signal transduction. Proteolytically cleaves MYOC at 'Arg-226' (PubMed:17650508).

VALIDATION IMAGES

Lane 1: A549 Cell lysates; Lane 2: SK-BR-3 Cell lysates; Lane 3: C2C12 Cell lysates; probed with CAPN2 (1C1) Monoclonal Antibody, unconjugated (bsm-51429M) at 1:1000 overnight at 4°C followed by a conjugated secondary antibody for 60 minutes at 37°C.


Paraformaldehyde-fixed, paraffin embedded human stomach tissue; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 15 minutes; Blocking buffer (3% BSA) at room temperature for 30min; Antibody incubation with CAPN2 (1C1) Monoclonal Antibody (bsm-51429M) at 1:25 for 1 hour at 37°C, followed by a conjugated secondary antibody for 20 minutes and DAB staining.


Flow cytometric analysis of U-87 MG cells using bsm-51429M CAPN2 Antibody (green) compared to mouse IgG2b isotype control (blue). bsm-51429M was diluted at 1:100 dilution. Alexa Fluor® 488 goat anti-mouse lgG at 1:400 dilution was used as the secondary antibody.
