Background:

Recognizes and binds the 7-methylguanosine-containing mRNA cap during an early step in the initiation of protein synthesis and facilitates ribosome binding by inducing the unwinding of the mRNAs secondary structures. Component of the CYFIP1-EIF4E-FMR1 complex which binds to the mRNA cap and mediates translational repression. In the CYFIP1-EIF4E-FMR1 complex this subunit mediates the binding to the mRNA cap.

VALIDATION IMAGES

Lane 1: Mouse spleen; Lane 2: Rat spleen cell lysate probed with eIF4E(S209) (2B1) Monoclonal Antibody (bsm-52152R) at 1:1000 overnight at 4˚C. Followed by a conjugated secondary antibody.


SH-SY-5Y cells were fixed in paraformaldehyde, permeabilized with 0.25% Triton X100/PBS and stained with eIF4E(S209) (2B1 ) Monoclonal Antibody (bsm-52152R) at 1:200 and incubated overnight at 4C, followed by secondary antibody incubation, DAPI staining of the nuclei and detection.


Lane 1: Mouse Pancreas tissue lysates; Lane 2:Rat Lymph node tissue lysates; Lane 3: Rat Cerebrum tissue lysates; Lane 4: Rat Pancreas tissue lysates ; Lane 5: Human Hela cell lysates;Lane 6: Human 293T cell lysates ;Lane 7: Human MCF-7 cell lysates; Lane 8: Human SH-SY5Y cell lysates probed with eIF4E(S209) (2B1) Monoclonal Antibody, Unconjugated (bsm-52152R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.


Paraformaldehyde-fixed, paraffin embedded Mouse brain; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with eIF4E(S209) (2B1) Monoclonal Antibody, Unconjugated (bsm-52152R) at 1:200 overnight at 4°C, DAB staining.
