Lane 1: Mouse Lung tissue lysates; Lane 2: Mouse Placenta tissue lysates; Lane 3: Mouse Testis tissue lysates; Lane 4: Rat Testis tissue lysates ; Lane 5: Human HepG2 cell lysates; Lane 6:Human MCF-7 cell lysates; Lane 7: Human U87MG cell lysates; Lane 8: Human SH-SY5Y cell lysates probed with EPAS1 (2G3) Monoclonal Antibody, Unconjugated (bsm-54278R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
The HepG2 (treated with CoCl2(100μM, 4h))(H) cells were fixed with 4% PFA (10 min at r.t.) and then permeabilized with 90% ice-cold methanol for 20 min at -20℃,the cells then were incubated in 5%BSA to block non-specific protein-protein interactions (30 min at r.t.). Cells stained with Primary Antibody for 30 min at room temperature,followed by secondary antibody incubation for 40 min at room temperature. Primary Antibody (green):Rabbit Anti-HIF-2 alpha antibody (bsm-54278R,1:100); Isotype Control (orange): Rabbit IgG (bs-0295P). Blank control (black): PBS. Acquisition of 20,000 events was performed.
